Sypherd, Paul S. (University of California, San Diego). Formation of ribosomes from precursor ribonucleoprotein particles. J. Bacteriol. 90:411-417. 1965.-Amino acid deprivation of a "relaxed" mutant leads to the accumulation of ribonucleoprotein particles. The possibility that these particles are equivalent to normal ribosome precursors was investigated. This was accomplished by determining the fate of such particles after the termination of starvation, and subsequent growth. It was found that both the nucleic acid and the protein moieties of the "relaxed" particles were converted to mature ribosomes during the recovery phase. From kinetic measurements with "relaxed" particles and normal ribosome precursors, it was shown that they were metabolically indistinguishable. It was concluded that the ribonucleoprotein "relaxed" particles accumulated by the relaxed mutant are immature ribosomes, and equivalent to ribosome precursors which occur under normal growth conditions.

THE general way of thinking about the pathway of expression of genetic information in eukaryotes, from DNA to protein via RNA, instills a protein-centric point of view, as if the protein is the real goal and the RNA is just the intermediate. In fact, most of the RNA in the cell is not mRNA but rather part of modular structures, RNA-protein machines, that are termed ribonucleoprotein (RNP) parti...
متن کاملRibonucleoprotein Particles in the Amphibian Oocyte Nucleus
Studies of the sedimentation properties of RNP(1) material from the nucleus of the amphibian oocyte have indicated (1) that there are few, if any, 78S ribosomes in the nucleus, (2) that there are smaller particles sedimenting at 50-55S and 30S, and (3) that the larger of these is the precursor of the 60S subunit of the cytoplasmic ribosomes. Although the nature of the 30S material is not comple...

There is much support for the view that the functional unit of protein synthesis is an aggregate of ribosomes held together by a strand of messenger RNA.1-6 The concept derives from the observation that radioactive amino acids are preferentially incorporated into ribosomal aggregates (termed polyribosomes, polysomes, or ergosomes) and that treatment of the ribosomal aggregates with small amount...

The in vitro assembly of Escherichia coli 50 S ribosomes from the ribonucleic acid and protein components of 50 S ribosomes was attempted. The two components were prepared by subjecting 50 S ribosomes to LiCl-urea treatment. The ribonucleic acid fraction contains 23 S RNA and 5 S RNA in equimolar amounts and is free from significant amounts of protein. The protein fraction is composed of all of...
